Deciphering bacteria associated with a pre-parasitic stage of the root-knot nematode Meloidogyne hapla in nemato-suppressive and nemato-conducive soils

چکیده
Nematode-suppressive soils are characterized by the ability of soil microbial communities to reduce populations plant-parasitic nematodes (PPN) either directly or inducing systemic resistance in plants. Various microorganisms have been recognized as antagonists PPN suppressive using culture-dependent and culture-independent methods. However, associations that form with nematode-conducive poorly studied. Here we drenched tomato rhizospheres suspensions from nine different followed their effects on plant growth root invasion infective second-stage juveniles (J2) northern root-knot nematode species Meloidogyne hapla . Based number invaded J2, four were determined nematode-suppressive, while five categorized nematode-conducive. To reveal bacteria attached J2 cuticle varying suppressiveness, incubated three conducive soils, analyzed J2-attached amplicon sequencing V3-V4 region 16S rRNA gene. Our results suggest origin had a major effect composition bacteria, highest bacterial abundance richness observed two soils. In addition, indicator sequence variants (ASVs) was associated but they very distinct profile. Further studies needed resolve complexity nemato-microbial interactions determine exact function nematode-attached role suppression protection. • Microbiomes uniquely affected J2. upon incubation Soil bacteria. Higher diversity found

Root-knot nematodes (RKN) are the most serious plant parasitic nematodes having a broad host range exceeding 2,000 plant species. Quercus brantii Lindl. and Q. infectoria Oliv are the most important woody species of Zagros forests in west of Iran where favors sub-Mediterranean climate. National Botanical Garden of Iran (NBGI) is scheduled to be the basic center for research and education of bot...
متن کاملThe cold tolerance of the northern root-knot nematode, Meloidogyne hapla
The northern root-knot nematode, Meloidogyne hapla, is one of the most important nematode pathogens occurring in cold regions. It is a sedentary, biotrophic parasites of plants and overwinter in the soil or in diseased roots. This study showed that the cold tolerance for the second-stage juveniles (J2) of M. hapla was moderate with the 50% survival temperature (S50) of -2.22°C and the fatal tem...
متن کاملImpact of Grapevine (Vitis vinifera) Varieties on Reproduction of the Northern Root-Knot Nematode (Meloidogyne hapla).
One of the most commonly encountered plant-parasitic nematodes in eastern Washington Vitis vinifera vineyards is Meloidogyne hapla; however, limited research exists on the impact of this nematode on V. vinifera. The objectives of this research were to determine the impact of M. hapla on Chardonnay and Cabernet Sauvignon vine establishment and to determine the host status of V. vinifera varietie...

Hirsutella rhossiliensis and Verticillium chlamydosporium infected second-stage juveniles (J2) and eggs of Meloidogyne hapla, respectively, in petri dishes and in organic soil in pots planted to lettuce in the greenhouse. In vitro, H. rhossiliensis produced 78 to 124 spores/infected J2 of M. hapla. The number of J2 in roots of lettuce seedlings decreased exponentially with increasing numbers of...
متن کاملMeloidogyne petuniae n. sp. (Nemata: Meloidogynidae), a Root-knot Nematode Parasitic on Petunia in Brazil.
Meloidogyne petuniae n. sp. is described and illustrated from specimens parasitic on petunia (Petunia hybrida L.) in Brasilia, Brazil. The perineal pattern of the female is elongate to ovoid with a high, squarish arch and widely spaced, coarse striae. The stylet of the female is 12.9-16.5 microm long and has three small, rounded knobs that are distinctly set off from the shaft. Each knob is mar...
